| Summary: | This study focused on detection of creatine kinase (CK) as a vital biomarker for diagnosing and monitoring various medical conditions, including myocardial infarction and muscle disorders. This study presents the development of a PAH-aptamer/AgNP-based colorimetric biosensor for the sensitive and specific detection of CK-MM. Silver nanoparticles (AgNPs) were synthesized via a bottom-up technique and functionalized with poly(allylamine hydrochloride) (PAH) and a thiolated CK-MM aptamer. The FTIR analysis indicated successful conjugation of PAH and aptamer to the AgNPs. The PAH-Aptamer/AgNP-based colorimetry assay showed a high sensitivity with a detection limit of 9 pg/mL and a wide linear detection range from 0.1 ng/mL to 104 ng/mL. Comparative studies with an ELISA kit demonstrated good agreement, with recovery rates ranging from 96.00 % to 99.10 % and relative standard deviations between 3.72 % and 4.78 %. The PAH-aptamer/AgNP-based biosensor offers a rapid, cost-effective, and practical alternative to traditional detection methods, making it a valuable tool for clinical diagnostics and monitoring physiological conditions in sports injury rehabilitation. This study underscores the potential of using PAH-aptamer/AgNPs in colorimetric sensing systems, providing a robust platform for early diagnosis and monitoring of CK-related conditions with high sensitivity, specificity, and practicality. |
