microRNA抑制黑色素瘤VEGF基因表达及对细胞增殖和 侵袭能力的影响
【目的】 研究外源性靶向microRNA对恶性黑色素瘤细胞血管内皮生长因子(VEGF)基因表达的抑制作用,以及对癌细胞增殖和侵袭迁移能力的影响?【方法】根据VEGF基因序列人工设计合成4对microRNA干扰片段,构建质粒真核表达载体,分别为:X-26-1-3?X-26-2n-1?X-26-3-2和X-26-4-4,测序分析鉴定插入序列的完整性;脂质体法将该四个重组表达载体转染黑色素瘤细胞株SKmel-28,RT-PCR检测转染细胞中VEGF基因的mRNA表达变化,Western Blot检测microRNA对VEGF蛋白表达水平的调控效应,明确抑制效果最为显著的microRNA干扰序列?MT...
