Extracellular vesicles, RNA sequencing, and bioinformatic analyses: Challenges, solutions, and recommendations
Published 2024-12-01“…Here, we address these gaps by describing the following points by (1) focusing on the large canopy of the EVs and particles (EVPs), which includes, but not limited to – exosomes and other large and small EVs, lipoproteins, exomeres/supermeres, mitochondrial‐derived vesicles, RNA binding proteins, and cell‐free DNA/RNA/proteins; (2) examining the potential functional roles and biogenesis of EVPs; (3) discussing various transcriptomic methods and technologies used in uncovering the cargoes of EVPs; (4) presenting a comprehensive list of RNA subtypes reported in EVPs; (5) describing different EV‐RNA databases and resources specific to EV‐RNA species; (6) reviewing established bioinformatics pipelines and novel strategies for reproducible EV transcriptomics analyses; (7) emphasizing the significant need for a gold standard approach in identifying EV‐RNAs across studies; (8) and finally, we highlight current challenges, discuss possible solutions, and present recommendations for robust and reproducible analyses of EVP‐associated small RNAs. …”

Establishment of the fingerprint of ganoderma lucidum spore powder and study on the anti-inflammatory activity of ganoderma lucidum spore oil
Published 2025-01-01“…[Methods] High Performance Liquid Chromatography and High Performance Thin-layer Chromatography techniques were used to analyze the chemical composition of Ganoderma lucidum spore powder from different origins; MTT method was used to determine the cell viability; ELISA was used to determine the RAW264.7 IL-1β, IL-6 and TNF-α levels in cell supernatant. [Results] The HPLC fingerprints were calibrated with a total of 12 peaks, of which peaks 1-5 were ganoderic acid C2, ganoderic acid G, ganoderic acid B, ganoderic acid A, ganoderic acid D, respectively; the current version of the Pharmacopoeia method of HPTLC showed that 10 batches of samples complied with the regulations, and the quality of raw materials of different origins was found to be similar to that of the HPLC fingerprints after 2 unfoldings by using trichloromethane-acetonitrile-methanol-formic acid (26∶4 ∶1∶1) as the unfolding agent. …”
