    “…【目的】 通过研究胆汁淤积性肝病不同阶段的肝脏病变65380;BDEC增殖及转化生长因β(TGF-β)的变化,以探讨胆汁淤积性肝病发生发展的可能机制65377;【方法】 ①以胆总管结扎(BDL)制作胆汁淤积性肝病大鼠模型,分为正常对照组(D0)65380;7天组(D7)及18天组(D18);②动态观察不同时间点大鼠肝脏病理变化,以Knodell组织学评分评价肝脏组织学改变;③自肝脏中分离BDEC,以实时定量PCR方法(RT-PCR)检测不同时间点肝脏组织以及分离所得BDEC中TGF-β表达的情况;④分析肝脏Knodell组织学评分与TGF-β1 mRNA表达水平间的相关性;⑤利用永生化的小鼠肝内胆管上皮细胞株(mIBEC),通过体外实验探讨TGF-β1对mIBEC增殖的作用65377; 【结果】 ①肝脏Knodell组织学评分随着淤胆时间延长而增加(P < 0.01),且胆管增殖程度随着淤胆时间延长而加重;②肝脏组织及BDEC中TGF-β165380;TGF-β2及TGF-β3的mRNA表达水平随着淤胆时间延长而显著上调;③肝脏Knodell组织学纤维化程度评分与不同时间点肝脏组织及BDEC中TGF-β1 mRNA表达水平间存在正相关(r = 0.9376,P < 0.05及r = 0.9682,P < 0.01)65377;④体外研究显示,TGF-β1可抑制mIBEC增殖65377;【结论】 ①BDL诱导的胆汁淤积性肝病中,肝脏损伤65380;胆管增殖程度及TGF-β mRNA表达水平均随淤胆时间延长而增加;②TGF-β1与BDEC的相互作用在BDL诱导的胆汁淤积性肝病发生发展中起着重要作用;③增生的BDEC中TGF-β1表达上调参与了BDL诱导的胆汁淤积性肝纤维化形成65377;…”
