    “…【目的】 研究槲皮素对耐甲氨蝶呤骨肉瘤细胞株U2-OS/MTX300的增殖抑制和诱导凋亡作用及相关机制【方法】 以不同浓度槲皮素及甲氨蝶呤处理U2-OS及U2-OS/MTX300采用四甲基偶氮唑蓝(MTT)法及平板克隆检测细胞增殖,流式细胞仪检测细胞周期,Hochest 33258 染色及流式细胞仪检测凋亡,Western blot法检测凋亡相关蛋白cleaved-PARP,Bax,Bak,Bcl-2及Bcl-XL表达【结果】 槲皮素可明显抑制耐甲氨蝶呤骨肉瘤细胞U2-OS/MTX300的增殖,72 h IC50值为(22 ± 4)μmol/L102550μmol/L可显著抑制U2-OS/MTX300克隆形成数量,且随剂量增加抑制作用明显增强槲皮素与甲氨蝶呤之间不存在交叉耐药性槲皮素能诱导U2-OS/MTX300细胞S期阻滞,并可诱导胞内产生典型凋亡小体,流式细胞仪可检测到明显凋亡其作用机制是通过上调Bax,Bak及下调Bcl-2表达,促进cytochrome C释放及诱发PARP等重要胞内蛋白断裂来实现【结论】 槲皮素可显著抑制耐甲氨蝶呤骨肉瘤细胞系U2-OS/MTX300细胞增殖并诱导其凋亡,线粒体途径可能是其诱导凋亡的重要机制…”
