    “…【目的】 探讨超顺磁氧化铁纳米颗粒(SPIO)标记对骨髓间充质干细胞生物学特性和多向分化潜能的影响以及体外磁共振(MRI)成像的可行性,为移植干细胞活体MRI成像提供实验基础65377; 【方法】 采用梯度密度离心法和贴壁法获取大鼠骨髓间充质干细胞(BMSC),采用25 μg/mL SPIO联合0.75 μg/mL多聚赖氨酸(PLL)标记BMSC,比较标记和未标记MSC细胞活力65380;增值65380;细胞周期65380;凋亡和成骨65380;成脂多向分化能力;应用1.5T MR对不同数量级细胞分别进行T1WI65380; T2WI和T2*WI序列扫描65377; 【结果】 普鲁士兰染色显示SPIO(25 μg Fe/mL, 48 h)对细胞的标记率接近100%,细胞活力65380;增殖65380;周期和凋亡检测,均显示SPIO标记细胞与未标记细胞间无统计学差异(P > 0.05);成骨65380;成脂诱导显示,BMSC和标记BMSC均具备成骨65380;成脂分化能力;对不同数量级的SPIO标记BMSC行MR扫描,T1WI65380; T2WI和T2*WI能检测到的最小数量级细胞分别为2 × 104, 1 × 104, 0.5 × 104, 呈低信号65377; 【结论】 25 μg/mL SPIO联合0.75 μg/mL PLL能有效标记BMSC,不影响BMSC的活力65380;增值65380;细胞周期65380;凋亡和多向分化能力,1.5T MR示踪标记细胞可行,与T1WI和T2WI序列相比,T2*WI最敏感65377;…”
