description | Objective:To explore the hepatic protection of Pien Tze Huang Gan Bao (GB) on chemical liver injury.Methods:Sixty SD rats were randomly divided into control group, model group, silymarin group, and GB-low dose group, GB-middle dose group, GB-high dose group with ten rats per group. The control group received olive oil (1 mL/kg) intraperitoneally, rest of the groups received CCl<sub>4</sub> (1 mL/kg) intraperitoneally, twice per week for 8 weeks. From the fourth to the eighth week, the model group and the control group of rats were orally administrated saline, while the silymarin group of rats were administrated silymarin (3.3 mg/kg), and GB-Low group, GB-middle group, and GB-high group of rats were administrated 150 mg/kg, 300 mg/kg, and 600 mg/kg of GB, respectively. Twenty-four hours after the last dose, MDA, SOD, GSH and TBARs were detected; the histopathological changes in liver were observed by HE staining, and CYP2E1 gene transcription was determined by Real-time PCR.Results:The contents of MDA and TBARs were decreased in both silymarin and GB treated group of rats and with increasing the activity of SOD and GSH (<italic>P</italic><0.05);Compared with model, silymain and GB ameliorated the liver injuried. Furthermore, silymarin and GB down-regulated the expression of CYP2E1 (<italic>P</italic><0.05).Conclusion:GB could ameliorate the chemical liver injury, and it maybe correlate with the anti-oxidant activity of GB. |
