description Autophagy is an effective protective mechanism against tissue degeneration and plays an important role in cell proliferation, differentiation and maturation. The main cells involved in bone me-tabolism include osteoblasts and osteoclasts, which play an important role in bone development and maintenance. It has been found that the level of autophagy is regulated by sirtuin1 (SIRT1) and mito-gen-activated protein kinase 8 (MAPK8)/forkhead box O3 (FOXO3) in osteoblasts. In osteoclasts, the level of autophagy is regulated mainly by Bcl-2 interacting coiled-coil protein 1 (Beclin-1), p62/sequestosome 1 (p62/SQSTM1), mammalian target of rapamycin (mTOR) and hypoxia-inducible factor-1α (HIF-1α). The main focus of this article is a discussion of the autophagy related signal trans-duction pathways in bone metabolism and an analysis of the regulation of autophagy in osteoblasts and osteoclasts.
