Summary:Cancer is among the leading causes of mortality across the globe. Despite the development of wide variety of oncotherapeutics, the effectiveness of oncotherapeutic drugs and anti-cancer bioactive compounds is frequently hindered by their low solubility and poor bioavailability, highlighting the need for efficient drug delivery system. Cyclodextrin (CD), a cyclic oligosaccharide produced by amylase hydrolysis of starch has emerged as promising candidates for drug delivery system due to their unique properties and low toxicity profile. This review comprehensively discussed the structure and properties of CD and its derivatives, as well as elucidated the synthesis and characterization of different types of CD-based drug delivery system including inclusion complex, nanoparticles, nanosponges, nanofibers, micelles, liposomes and hydrogels. The application of CD-based drug delivery system for oncotherapeutics was discussed and a forward-looking perspective on the challenges and prospects of CD-based drug delivery system demonstrated.
