Summary:This article analyze the important and the role of two middle cities - Marabá and Santarém -in the context of the spatial transformations in Brazilian Amazon, which are responsible for define new urban-regionals centralities and a complex territorial structure. Based on bibliographical and documentary research it analyzes the historical and geographical factors responsible for the leadership role these cities in the regional context. Moreover, we recognize and discuss through of primary and secondary data elements the urban centrality exerted for Marabá e Santarém in their respective sub-regions, Southeast and West Pará’s State. This centrality is considered in its economic dimension and mainly in its political dimension in view of the proposal of territorial fragmentation for the creation of the two new federation States - Carajás and Tapajós.
