Protective Mechanism of Electroacupuncture Combined with Enriched Rehabilitative Training on Hippocampal Neurons in Rats with Cerebral Ischemia-Reperfusion Injury

Summary: | Objective:To observe the effects of electroacupuncture combined with enriched rehabilitative training on endoplasmic reticulum stress (ERS) -associated proteins in rats with cerebral ischemia-reperfusion injury, and to investigate the protective mechanism of that in relation to the hippocampal neurons.Methods:A total of 108 enrolled Sprague-Dawley (SD) rats were divided into normal group, sham surgery group, model group, electroacupuncture group (EA group), enriched rehabilitative training group (RT group), and electroacupuncture combined with enriched rehabilitative training group (EA+RT group) according to the random number table method. Then cerebral ischemia-reperfusion models were established by modified thread thrombus method. After the successful modeling in the EA group and EA+RT group, electroacupuncture was performed at Baihui (GV20) and Shenting (GV24) acupoints. For the rats in the RT group and EA+RT group, an enriched environment was set up and they were involved in rehabilitative activities. The learning and memory abilities of rats were assessed by water maze test. The cerebral infarction volume was observed via TTC staining. The neuronal apoptotic index was determined by TUNEL method. Western blot was used to detect the expression of protein kinase-like endoplasmic reticulum kinase (PERK), CCAAT/enhancer binding protein homologous protein (CHOP), B-cell lymphoma-2(Bcl-2), Bcl-2 associated X-protein (Bax), protein kinase B(Akt), and phosphory lated protein kinase B(p-Akt).Results:Compared withthenormalgroupandshamgroup, learning and memory ability of rats in the model group were decreased (<italic>P</italic>< 0.01), and cerebral infarction volume, apoptosis index, protein expression of PERK, CHOP, Bcl-2 and Bax in hippocampus of rats in the model group were significantly increased (<italic>P</italic>< 0.01). Compared with the model group, learning and memory ability of the rats in the three treatment groups were increased (<italic>P</italic>< 0.01), cerebral infarction volume, apoptosis index, protein expression of PERK, CHOP and Bax were significantly decreased (<italic>P</italic>< 0.01), and protein expression of Bcl-2 and p-Akt level were increased (<italic>P</italic>< 0.01). Compared with the EA group and RT group, the EA+RT group showed stronger learning and memory ability (<italic>P</italic>< 0.01), lower cerebral infarction volume, apoptosis index, protein expression of PERK, CHOP and Bax (<italic>P</italic>< 0.05), higher Bcl-2 protein expression and p-Akt level (<italic>P</italic>< 0.01). Comparision between the normal group and sham group, between the EA group and RT group showed no statistical significance (<italic>P</italic>> 0.05).Conclusion:Electroacupuncture combined with enriched rehabilitation training has a good therapeutic effect on cerebral ischemia-reperfusion injury, which is better than the single use of electroacupuncture or rich rehabilitation training. Its effect may inhibit the release and activation of ERS-associated proteins PERK, CHOP and Bax, up-regulate the expression of anti-apoptotic protein Bcl-2 and p-Akt, interfere with ERS-mediated apoptosis pathway and reduce the apoptosis rate, so that the cerebral nerve can be protected. |
