| Summary: | The binding of acetochor, pretilachor, butachlor and metolachlor to urease in aqueous solution was studied using UV difference spectrum and fluorescence spectrum methods. According to the UV difference spectrum curve obtained, the UV difference adsorption AA showed rapid change when herbicide concentrations ranged from 0.0 to 1.6 μmol/L. Red shift occurred in the UV difference spectra of urease and absorbance-peaks were lower. It was shown that the herbicides quenched the urease fluorescence mainly through a static quenching process. The binding constant K and the number of binding site n were calculated according to the fluorescence quenching results. In case of acetochor, K=1.17×103 L/mol and n was 0.81; to pretilachor, K=1.46×102 L/mol and n was 0.67; whereas for butachlor, K=29×101 L/mol and n was 0.50; and for metolachlor, K=1.49×103 L/mol and n was 0.84. |
